Induction of apoptosis by extract of Phlox paniculata L in KB human oral cancer cells

Abstract
Purpose: Phlox paniculata L, belonging to the Polemoniaceae family, is widely planted for ornamental purposes in Korea. Here, we investigated the molecular basis underlying the antitumor effects of Phlox paniculata L.

Materials and Methods: For this, we analyzed the effects of this plant on the proliferation and expression of cell apoptosisrelated molecules in KB human oral cancer cells. The proliferation test was conducted by MTT assay. Treatments with Phlox paniculata L extract dramatically reduced cell viabilities in a dose- and time-dependent manner. In addition, typical apoptosis patterns were detected by DNA fragmentation assay and Western blot analysis.

Results and Conclusion: Western blot data revealed that activation of caspases-3 and -7 increased after 24 hours and 12 hours of treatment, respectively. We also confirmed DNA fragmentation upon treatment with Phlox paniculata L extract in KB human oral cancer cells. Collectively, these results suggest that Phlox paniculata L may have beneficial effects on the proliferation of oral cancer cells through activation of apoptosis.
